Carbonic anhydrase IX (CA9/CAIX), also known as MN protein or G250 antigen, is a transmembrane zinc metalloenzyme that catalyzes the reversible hydration of carbon dioxide to bicarbonate and protons. Primarily regulated by hypoxia-inducible factor 1-alpha (HIF-1α), it maintains intracellular pH homeostasis while acidifying the extracellular tumor microenvironment, thereby promoting cancer cell survival, invasion, and metastasis. CA9 is functionally linked to PI3K/AKT and ERK signaling pathways, with overexpression observed in clear cell renal cell carcinoma (ccRCC), triple-negative breast cancer, and colorectal adenocarcinoma. Its tumor-specific expression has positioned CA9 as both a diagnostic biomarker and therapeutic target. Clinical-stage agents include the small-molecule inhibitor SLC-0111 (Phase II) targeting CA9 enzymatic activity, and antibody-based therapies like ⁸⁹Zr-labeled girentuximab for PET imaging. CA9-targeted chimeric antigen receptor (CAR) T-cell therapies are under preclinical evaluation for ccRCC management.

IHC image of CSB-RA614990A0HU diluted at 1:118 and staining in paraffin-embedded human gastric cancer performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4℃ overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
