ITGA9

Integrin alpha-9 (ITGA9), also known as α9 integrin, forms a heterodimer with the β1 subunit (ITGB1) to constitute integrin α9β1. This transmembrane receptor mediates cell-cell and cell-extracellular matrix interactions through binding to ligands such as VCAM-1, osteopontin, and tenascin-C. It plays pivotal roles in cellular adhesion, migration, and mechanotransduction, primarily via modulating Rho GTPase activity and activating focal adhesion kinase (FAK)-mediated signaling. ITGA9 is functionally implicated in the TGF-β/Smad pathway and lymphatic development. Dysregulation of ITGA9 has been linked to pathological processes including cancer metastasis, chronic obstructive pulmonary disease (COPD), and rheumatoid arthritis. Its overexpression in tumors correlates with enhanced invasive potential and poor prognosis.
Current therapeutic strategies targeting ITGA9 focus on monoclonal antibodies and small-molecule inhibitors to disrupt its ligand-binding interface or downstream signaling. Preclinical studies demonstrate that α9β1 blockade reduces leukocyte infiltration in inflammatory models and inhibits tumor angiogenesis. A humanized anti-ITGA9 antibody (ASKB90) has entered Phase I trials for solid tumors, while peptide-based inhibitors show efficacy in experimental pulmonary fibrosis models. Challenges remain in achieving tissue-specific targeting due to ITGA9's physiological roles in lymphatic valve formation and wound healing.
