Integrin alpha-V (ITGAV), also known as CD51 or VTNR, is a transmembrane receptor that primarily pairs with β-subunits (e.g., β3, β5, β6) to form heterodimeric complexes. These complexes bind extracellular matrix (ECM) ligands such as fibronectin and vitronectin, facilitating cell adhesion, migration, and bidirectional signaling. ITGAV is critical in pathological processes like tumor angiogenesis, epithelial-mesenchymal transition (EMT), and fibrosis. Dysregulation of ITGAV is implicated in aggressive cancers (e.g., glioblastoma, breast cancer), chronic fibrotic disorders (e.g., liver cirrhosis), and osteoporosis, often correlating with poor prognosis.
Targeting ITGAV has emerged as a therapeutic strategy. The cyclic RGD peptide Cilengitide, an αvβ3/αvβ5 integrin antagonist, advanced to Phase III trials for glioblastoma but failed to meet efficacy endpoints. Current efforts focus on antibody-drug conjugates (ADCs) and small-molecule inhibitors to block ITGAV-mediated TGF-β activation or ECM remodeling. Preclinical models highlight synergies with immune checkpoint inhibitors, while novel αvβ6-specific agents show promise in treating pulmonary fibrosis. Challenges remain in optimizing selectivity and overcoming compensatory pathways in clinical translation.

IHC image of CSB-RA279866A0HU diluted at 1:100 and staining in paraffin-embedded human placenta tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT.
