This tag-free recombinant rhesus macaque IL-1β, expressed in *E. coli* as the full-length mature protein (117–269 aa), demonstrates exceptional signaling potency with an ED50 below 10 pg/mL in D10.G4.1 proliferation assays, corresponding to a specific activity exceeding 1.0 × 10⁸ IU/mg. Endotoxin levels below 1.0 EU/μg ensure that downstream immune cell responses—including NF-κB pathway activation, T-cell proliferation, and macrophage differentiation assays—reflect genuine IL-1β signaling rather than LPS-driven artifacts. With purity above 98% by SDS-PAGE and stringent endotoxin control, this protein provides a suitable basis for use as a reference standard in cytokine detection assays, a functional immunogen for antibody development, and a reliable tool in nonhuman primate *in vivo* inflammation models.
