Description
PTK7 functions as a pseudokinase receptor implicated in Wnt signaling and cancer progression, yet its catalytically inactive kinase domain complicates the production of conformationally authentic extracellular fragments for antibody development and ligand discovery. This mammalian-expressed construct spanning residues 31–704 captures the complete extracellular region, preserving native glycosylation and disulfide architecture essential for antibody epitope presentation. Functional ELISA validation demonstrates specific binding to an anti-PTK7 recombinant antibody with an EC50 of 0.37–0.66 ng/mL, confirming that the protein adopts a conformation suitable for therapeutic antibody epitope mapping, blocking antibody screening, and competitive inhibition assays targeting PTK7-mediated oncogenic pathways. The protein meets purity thresholds exceeding 95% by SDS-PAGE and endotoxin levels below 1.0 EU/μg, aligning with standards expected in surface plasmon resonance affinity characterization and biolayer interferometry experiments for drug discovery programs focused on PTK7 as a cancer target.
