The VLPs are expressed from human 293 cells (HEK293).Mix the sample gently by repeatedly pipetting it up and down. Do not vortex.Repeated freezing and thawing is not recommended.Store the protein at -20°C/-80°C upon receiving it, and ensure to avoid repeated freezing and thawing, otherwise, it will affect the protein activity. The immunization strategy should be optimized (antigen dose, regimen and adjuvant).

Description

Claudin-18.2 presents a unique challenge for recombinant production because its four-transmembrane topology demands a native membrane context to preserve conformational epitopes critical for antibody recognition. This fluorescent VLP-displayed construct presents full-length human CLDN18.2 (aa 1–261, isoform A2) with a C-terminal mCherry tag, enabling direct visualization in flow cytometry-based antibody screening and cell-based binding assays. Functional ELISA confirms that immobilized CLDN18.2 VLPs bind an anti-CLDN18.2 recombinant antibody with an EC50 of 5.1–8.5 ng/mL, demonstrating preserved extracellular loop conformation suitable for antibody development and validation, therapeutic antibody benchmarking, and tumor metastasis research targeting CLDN18.2-positive cancers. The mammalian-expressed VLP format maintains native folding and membrane orientation while endotoxin levels below 1.0 EU/μg satisfy criteria appropriate for immune cell trafficking studies and integrin-ligand binding assays requiring low background activation.
