PD-1 is a critical immune checkpoint receptor whose extracellular domain mediates interactions with PD-L1 and therapeutic antibodies targeting the PD-1/PD-L1 axis. This recombinant human PD-1 construct covers the ligand-binding extracellular domain (aa 25–167) and is expressed in mammalian cells to preserve native glycosylation and conformational integrity essential for accurate binding studies. Functional ELISA validation demonstrates robust activity, with EC50 values of 6.087–7.854 ng/ml against an anti-PD-1 recombinant antibody and 9.713–12.39 ng/ml against Nivolumab, confirming suitability for competitive blocking antibody screening, therapeutic antibody epitope mapping, and affinity characterization by SPR or BLI.
