Description

TNF-α drives apoptosis, inflammation, and immune cell activation through TNFR1 and TNFR2 signaling, making precise control of its bioactivity essential for mechanistic studies in cancer and inflammatory disease models. This mammalian-expressed rat TNF (aa 80–235) demonstrates potent cytotoxic activity with an ED50 of 7.7–15.5 pg/mL in L-929 fibroblast assays and binds human TNFRSF1A with an EC50 of 6.2–7.5 ng/mL and a KD of 0.139 nM, confirming receptor engagement at physiologically relevant concentrations suitable for NF-κB and MAPK pathway activation studies, apoptosis assays in tumor cell lines, and in vivo inflammation models. The endotoxin level below 1.0 EU/μg prevents LPS-driven artifacts that confound interpretation in macrophage, dendritic cell, and primary immune cell experiments where TNF and TLR4 pathways converge. Purity exceeding 95% supports its use as a coating antigen in ELISA development and as a reference standard in cytokine detection platforms.

Cytokine that binds to TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. It is mainly secreted by macrophages and can induce cell death of certain tumor cell lines. It is potent pyrogen causing fever by direct action or by stimulation of interleukin-1 secretion and is implicated in the induction of cachexia, Under certain conditions it can stimulate cell proliferation and induce cell differentiation. Induces insulin resistance in adipocytes via inhibition of insulin-induced IRS1 tyrosine phosphorylation and insulin-induced glucose uptake. Induces GKAP42 protein degradation in adipocytes which is partially responsible for TNF-induced insulin resistance. Plays a role in angiogenesis by inducing VEGF production synergistically with IL1B and IL6.; The TNF intracellular domain (ICD) form induces IL12 production in dendritic cells.
