HTR4

The 5-hydroxytryptamine receptor 4 (HTR4), commonly known as 5-HT4R, is a G-protein coupled receptor (GPCR) that signals via Gs proteins. Upon activation by serotonin, it stimulates adenylate cyclase to increase intracellular cyclic adenosine monophosphate (cAMP) levels, activating the cAMP-dependent protein kinase A (PKA) pathway. This mechanism regulates neurotransmitter release (e.g., acetylcholine in the gastrointestinal tract) and neuronal excitability.
HTR4 dysregulation links to gastrointestinal disorders like irritable bowel syndrome with constipation (IBS-C) due to its role in promoting intestinal motility. Neurologically, it modulates cognitive function, with implications for Alzheimer’s disease. Approved drugs include prucalopride, an HTR4 agonist for IBS-C, while investigational agents target cognitive impairment by enhancing cholinergic signaling.

Immunofluorescence staining of MCF-7 cells with CSB-PA010895LA01HU at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
