STEAP1

STEAP1, alternatively known as prostate transmembrane epithelial antigen 1, is a cell surface antigen induced by androgens and highly expressed in prostate cancer. It consists of six transmembrane helices, encompassing 339 amino acids, with intracellular N- and C-terminal domains linked by three extracellular loops and two intracellular loops. Distinguishing from other STEAPs, STEAP1 lacks the N-terminal NADPH-oxidoreductase domain, thus not independently exhibiting Fe3+ or Cu2+ reductase activity. However, its C-terminal domain contains a heme b cofactor. Additionally, STEAP1 is also highly expressed in Ewing's sarcoma (EWS) and is regulated by the EWS-FLI1 fusion protein, the primary oncogenic driver of this malignancy. Functionally, overexpression of STEAP1 is associated with malignant prostate tissue but is expressed at lower levels in normal prostate tissue, making it an ideal therapeutic target and potentially serving as a diagnostic and prognostic biomarker. Notably, STEAP1 also plays a significant role in lung cancer angiogenesis, a process crucial for solid tumor growth, which depends primarily on tumor cells and tumor vascular endothelial cells. The discovery of STEAP1's role in lung cancer angiogenesis offers novel insights for the treatment of this disease.
